为提高Western Blotting结果的可靠性,以蛋白溶出率和凝胶电泳蛋白条带完整性为指标,比较提取液种类、研磨方式、酶抑制剂种类及其体积分数和提取时间对南美白对虾肝胰腺蛋白提取效果的影响。采用优化后的提取方法获得高质量蛋白样品,并采用Western Blotting法分析无水环境胁迫后南美白对虾肝胰腺组织中细胞凋亡信号通路相关蛋白的表达水平。结果表明:RIPA裂解液作为提取溶剂所得的蛋白溶出率高于水提和磷酸盐缓冲液,电动匀浆和液氮研磨所得蛋白条带更完整,4%蛋白酶磷酸酶混合抑制剂能有效抑制肝胰腺内源酶引起的蛋白降解;采用Western Blotting法分析无水保活期间南美白对虾肝胰腺蛋白,发现低温诱导休眠的同时会引起细胞轻微凋亡,且凋亡水平呈应激时间依赖性增加,环境胁迫解除后有所回调。 相似文献

The demand for food production has been constantly increasing due to rising population. In developed countries, for example, the emergence of regional production of old grains that are rarely utilized, along with the production of commonly consumed grains, has gained importance in recent years. These grains, known collectively as ancient or heirloom grains, have offered both farmers and consumers novel ways of cultivation and products with interesting taste, characteristics and nutritional value. Among the 30 000 plant species known, only five cereals currently provide more than 50% of the world's energy intake – bread wheat (Triticum aestivum), rice (Oryza sativa), sorghum (Sorghum bicolor), millets (Panicum sp.) and maize (Zea mays). The excessive utilization of these selected species has a great potential to cause genetic losses and difficulty in bridging future agricultural demands. Teff (Eragrostis tef), an ancient grain extensively cultivated in countries like Eritrea and Ethiopia, provides promising alternatives for new food uses since its nutritional value is significantly higher than most others cereal grains. The absence of gluten allows flexibility in food utilization since it can be directly substituted to gluten-containing products. The grain also offers an excellent balance of essential amino acids and minerals, which can fulfil the recommended daily intake and eliminates the need for fortification and enrichment. This review provides a general overview of the physical properties and nutritional composition of teff grains related to processing and applications in the food and feed industries. The current status of teff utilization, as well as the challenges in production and commercialization, and future opportunities is presented and discussed. 相似文献

Dynamic control of the absorption frequency and intensity of metamaterial absorbers has attracted considerable attention,and many kinds of tunable metamaterial absorbers have been proposed.Unfortunately,due to the integration of separate resonant unit and tunable unit,these designed metamaterial absorbers suffer from complex structure and low sensitivity.We numerically and experimentally demonstrate a tunable metamaterial absorber composed of artificial dielectric atoms as both resonant and tunable unit arrayed periodically in the background matrix on the metallic plate.Polarization insensitive and wide incident angle absorption band with simulated and experimental absorptivity of 99%and 96%at 9.65 GHz are achieved at room temperature.The absorption frequency can be gradually modulated by temperature,however,the absorption intensity at working frequency remains near unity.The dielectric atoms based tunable metamaterial absorbers with simple structure have potential applications as tempe rature sensors and frequency selective thermal emitters. 相似文献

以黏胶短纤维和再生植物纤维为主要原料,采用短纤维干法成网-针刺加固工艺,在针刺密度为60~160针/cm2、针刺深度为3.5~ 12.0 mm、针刺频率为720~ 940次/min的条件下,制备单位面积质量在196~979g/m2、厚度在3.6~ 14.4 mm、再生植物纤维质量分数在39.6% ~ 70.9%的针刺复合生态土工布,用于植生护坡.性能测试和植物种植试验结果表明:针刺复合生态土工布具有较好的力学性能和吸水保水性能,且对植物的生长具有良好的促进作用;随着针刺复合生态土工布单位面积质量的增加,其力学性能增加,吸水保水性能变好,发芽率、植株高度和植株质量也均呈现出增加的趋势. 相似文献

The effects of La2O3–Al2O3–SiO2 addition on the thermal conductivity, coefficient of thermal expansion (CTE), Young's modulus and cyclic thermal shock resistance of hot-pressed h-BN composite ceramics were investigated. The samples were heated to 1000 °C and then quenched to room temperature with 1–50 cycles, and the residual flexural strength was used to evaluate cyclic thermal shock resistance. h-BN composite ceramics containing 10 vol% La2O3–Al2O3 and 20 vol% SiO2 addition exhibited the highest flexural strength, thermal conductivity and relatively low CTE, which were beneficial to the excellent thermal shock resistance. In addition, the viscous amorphous phase of ternary La2O3–Al2O3–SiO2 system could accommodate and relax thermal stress contributing to the high thermal shock resistance. Therefore, the residual flexural strength still maintained the value of 234.3 MPa (86.9% of initial strength) after 50 cycles of thermal shock. 相似文献

Formation of SiC whiskers/leucite-based ceramic composites from low temperature hardening geopolymer

In this study, SiC whiskers (SCWS) reinforced geopolymer composites (SCWS/KGP) and their ceramic products (SCWS/leucite) were prepared, and effects of SiC whiskers contents on the microstructure and flexural strength of the SCWS/KGP and SCWS/leucite composites were investigated. The results show that the whisker addition has little influence on both phase composition and thermal shrinkage of the KGP composites, but a suitable content of whisker will result in the improved flexural strength, and when the SCWS content is 2 wt%, flexural strength of the SCWS/KGP composite is enhanced by 95% compared with the neat geopolymer. The flexural strength of the composites can be further enhanced significantly after the composites being treated at 1100 °C and 1200 °C and flexural strength of the composite with SCWS content of 2 wt% was 107% and 125% higher than the untreated counterpart, respectively. The increase in flexural strength of the composites should be attributed to the strong leucite formation, whisker debonding and pulling out from matrix during the fracturing process based on the good interfacial bonding state between whisker and leucite matrix. 相似文献

氨基甲酸乙酯(EC)是发酵食品(面包、酸奶、大豆奶酪等)和酒精饮料(葡萄酒、清酒、黄酒等)在发酵过程中,含氮化合物不完全代谢产生的副产物,与乙醇自发反应产生的,对人体具有潜在遗传毒性和致癌性。黄酒是我国传统而独特的发酵酒,因独特的风味和较高的营养价值,而深受消费者的喜爱。然而,在传统黄酒中,氨基甲酸乙酯含量显著高于其它酒精饮料,对消费者健康构成潜在的安全隐患。降低黄酒中氨基甲酸乙酯含量对黄酒产业的可持续发展具有重要意义。本文综述尿素、瓜氨酸及精氨酸分别在酿酒酵母及乳酸菌细胞中代谢调控机制,以及黄酒酿造过程中EC消减的方法。 相似文献
